Enterococcus By Richard Guilford Enterococcus Enterococcus, or more appropriately enterococci, are a family of bacteria in the division firmicutes (meaning strong skin and referring to the cell wall) and the class bacilli (which refers to its rod like shape). They look just like streptococcus bacteria from physical characteristics alone. In fact, members of the genus Enterococcus were classified as Group D Streptococcus until 1984 when genomic DNA analysis showed that a separate genus classification would be appropriate. Enterococci are part of the normal intestinal flora (good bacteria) of humans and animals but are also important pathogens responsible for serious infections. They are considered gram-positive, meaning they stain dark blue or violet. They also mostly occur in pairs or short chains. Enterococci are facultative anaerobic organisms, meaning they can thrive in both oxygen rich and oxygen poor environments. They do not form spores as many bacteria do, meaning they do not have a reproductive structure that is adapted for dispersal and survival for extended periods of time in unfavorable conditions. Nevertheless, they are tolerant of a wide range of environmental conditions: such as extreme temperature (10-45 °C), pH (4. 5-10. 0) and high sodium chloride concentration. In bodies of water, the acceptable level of contamination is very low, for example in the state of Hawaii, with among the strictest tolerances in the United States, the limit for water off its beaches is 7 colony-forming units per 100 ml of water. Anything above that, the state may post warnings to stay out of the ocean. In 2004, Enterococcus spp. took the place of fecal coliform as the new federal standard for water quality at public beaches. It is believed to provide a higher correlation than fecal coliform with many of the human pathogens often found in city sewage (eww right? ). There are many strains of enterococcus. Important clinical infections caused by Enterococcus include urinary tract infections, bacteremia, bacterial endocarditis, diverticulitis, and meningitis. Sensitive strains of these bacteria can be treated with ampicillin and vancomycin, two well known and often used antibiotics. An important feature of enterococcus is the high level of intrinsic antibiotic resistance, meaning its inherent ability to ithstand some of the more common antibiotics. Some enterococci are intrinsically resistant to beta-lactam-based antibiotics (those include some penicillins and virtually all cephalosporins) as well as many aminoglycosides (an amino-sugar that can sometimes function as an antibiotic). In the last two decades, particularly virulent strains of enterococcus that are resistant to vancomycin (vancomycin-resistant E nterococcus, or VRE) have emerged in nosocomial (infections caught in hospitals while care is being given) infections of hospitalized patients especially in the US. Other developed countries such as the UK have been spared this epidemic, and, in 2005, Singapore managed to halt an epidemic of VRE. VRE may be treated with quinupristin/dalfopristin (a special antibiotic developed to fight VRE) or Synercid with response rates of approximately 70%. Enterococcus avium is a species that is most commonly found in birds. Rarely, it is also a cause of infection in humans, and in such cases, may be vancomycin-reistant. It is referred to as VREA. VREA cases in humans have been successfully treated with linezolid (a synthetic antibiotic). Enterococcus durans and enterococcus faecalis are very similar. They are often mistaken for one another in determining infections. They inhabit the gastrointestinal tracts of humans and other mammals. It is among the main parts of some probiotic food supplements. Like other species in the genus enterococcus, E. faecalis can cause life-threatening infections in humans, especially in the nosocomial environment. The naturally high levels of antibiotic resistance found in E. faecalis contribute to its ability to infect. E. faecalis has been frequently found in root canal-treated teeth in prevalence values ranging from 30% to 90% of the cases. Root canal-treated teeth are about nine times more likely to have E. faecalis than cases of primary infections. Enterococcus faecium is an alpha hemolitic or nonhemolitic, bacterium. It is used as a probiotic in animals. It can be commensal (a harmlessly coexisting organism) in the human intestine, but it may also be pathogenic, causing diseases such as neonatal meningitis. Enterococcal meningitis is also a rare complication of neurosurgery. It often requires treatment with IV or intrathecal (something introduced into or occurring in the space under the arachnoid membrane of the brain or spinal cord) vancomycin, yet it is uncertain whether its use has any impact on outcome. The removal of any neurological devices is an important part of the management of these infections. Enterococcus solitarius and Enterococcus gallinarum are two more species of enterococcus. The genus Enterococcus includes more than 17 species, but only a few cause clinical infections in humans. Infection control programs were created three decades ago to control antibiotic-resistant nosocomial infections, but there has been little evidence of control in most facilities. After long, steady increases of MRSA and VRE infections in hospitals, the Society for Healthcare Epidemiology of America (SHEA) Board of Directors made reducing antibiotic-resistant infections a strategic SHEA goal in January 2000. After 2 more years without improvement, a SHEA task force was appointed to draft an evidence-based guideline on preventing nosocomial transmission of pathogens, focusing on the two considered most out of control: MRSA and VRE. Medline searches were conducted spanning 1966 to 2002. Many interpretations of unpublished studies providing sufficient data were included. Frequent antibiotics in healthcare settings provided a small advantage for resistant strands, but patients with MRSA or VRE usually acquire it through spread. The CDC has long-recommended contact precautions for patients colonized or infected with these pathogens. Most facilities have required this as policy, but have not actively identified infected patients with surveillance cultures (lab tests performed to determine possible abnormalities or infection) and tests, leaving most infected patients undetected and not isolated. Many studies have shown control of endemic (exclusively native to a place) and/or epidemic (everywhere) MRSA and VRE infections using surveillance cultures and contact precautions. Active surveillance cultures are essential to identify the infected to prevent spread of MRSA and VRE infections and make control possible using the CDC’s long-recommended contact precautions. Testing for enterococci can be tricky. Enterococci have been recognized to be of fecal origin since the beginning of this century. The usual ecological home for the Enterococcus species is the intestines of humans and other animals. However, enterococci are ubiquitous (present everywhere) and can be found free-living in soil, on plants, or in dairy products. Enterococcus comprises gram-positive cocci that are catalase (a common enzyme found in nearly all living organisms that are exposed to oxygen, where it functions to catalyze the decomposition of hydrogen peroxide to water and oxygen) negative, usually facultative anaerobic acteria. There is clear evidence of the genotypic (concerning the genetic makeup) identity of Enterococcus, based on molecular studies. Enterococci have also been related to human diseases, becoming established as major nosocomial pathogens. The isolation of strains resistant to many antibiotics has become important in public health concerns. In addition, Enter ococcus and Streptococcus have been proposed as indicators of fecal contamination in water because of their high abundance in feces and their long survival in the environment. Although the ratio of fecal coliforms to fecal streptococci has been ruled out as an indicator, the identi? cation of species associated with a given environment or host might provide additional information about the origin and the source of fecal contamination. The ability of enterococci to grow under particular conditions is widely used in their selective identification. This characteristic allows the detection and enumeration of enterococci with a selective medium (template used to grow bacteria) M-enterococcus agar or KF streptococcus agar, and by using bile-esculin-azide agar (another medium) as a further test for con? mation. Although this approach can distinguish Enterococci from other bacterial species, some may be identi? ed wrongly. The use of these media can hurt either selectivity (choices of bacteria found) or productivity (ability for bacteria to grow). Even though this approach was to identify enterococci species, it is unsuitable for the detection of certain enterococ ci species because they do not grow on these media. In addition, other bacterial species such as Streptococcus are able to grow on the media, presenting results similar to those of Enterococci. Enterococci are diverse and enigmatic. There are many strains and many complications that can arise from them. They are both harmless and deadly, useful and destructive. They can be used to aid digestion, or feared in a hospital setting. They are indeed a medical paradox. With continued study, we hope to learn from and understand these bacteria if not to aid us in our expansion of knowledge, then to rid us of a potentially critical threat in our hospital settings. Writers block has always been an issue for not only authors but also for average Joes like me. It can be difficult to do any kind of writing when you are chained by your own mind. This is an immense issue because authors cannot develop ideas to put them into action on the page. Although there are many ways that writers do away with writers block, the best solution for is to take a pen to paper and just start letting ideas flow randomly onto the page so that your mind can break free. This solution is called free writing.Free writing is not only good for unblocking your mind, but also to keep your thoughts on the page so that you can keep your soul at peace. The idea behind free writing is that you must keep your pen to paper the whole duration of the writing session. Usually free write sessions are short, to get thoughts flowing before working on traditional piece. If you are not able to come up with a next word just write â€Å"blah, blah, blah† until something clicks. This ma kes it so that the writer must not pause to think of his next word, which keeps this form of writing completely free from thought interruption making it all from your heart.Traditional writing is when you have structure behind the piece of work and it must comply with standard grammar, spelling, and punctuation. This can impede with natural creativity by making a maze that you must find your way through in order to complete your piece of work. So many authors have trouble expressingtheir ideas and thoughts on a page when society tells them that they have to oblige by these rules. An example of the loops a writer has to jump through in order to complete their work is the assignment sheet of guidelines for this paper. This document goes over the â€Å"correct† way to write a problem-reducing essay.The grade you receive on this essay is determined by how well you can follow these guidelines. The truly great thing with free writing is that you can write in any style you choose, finding the best way to get thoughts stirring and on the page.As you can see in the work above, I have chosen poetry to get my thoughts on the page and this is the best way for me to express myself. Any one person can have a different way of expression in writing. The limitations brought by society can challenge this expression, but by free writing you can open up and do away with writers blo ck. Contemporary societies Essay The art of literature in contemporary societies has always led to more critical discussions between man and himself and between man and his neighbor. Literature pushes us to rethink our stance on normal societal norms and beliefs. Sula is such a work of art. It is the based on two women living at the Bottom in Ohio, a predominantly black community. Helen Wright is a socially conscious and quite a conservative woman. She has one daughter; Nel. Hannah Peace a beautiful flamboyant and a woman with many men at the Bottom is the mother of Sula. Sula is raised with Eva Peace, her grandmother who does not stifle her granddaughters freedom unlike Nel who comes from a restrictive household. Nel and Sula develop quite an inseparable and good friendship, a friendship that does not settle well with Helen who possesses grave misgivings due to the reputation of Sulas mother however she does not object openly after her realization that Sula is a very polite house guest. After High School these two great friends diverge ways for 10 years. Sula goes to college where she experiences a new wave of sexual promiscuity much like her mother. Nel On the other hand is married by a Bottoms resident, Jude Green. Their separation last for period of three years before Sula comes back from the college and their relationship resumes without any hindrance. However, this relationship is cut short when Nel learns of the relationship between Sula and Jude. This affair ends both the relationship between Jude and Nel as well as that of Nel as Sula who leaves the bottom for three years. Nel is forced to raise her two children alone and she doesnt communicate with Sula for the three years that she goes away. Their next meeting occurs when Sula is terribly sick and almost dying. Their last conversation before Sula dies constitutes the literary wealth of this masterpiece; it is the discussion between good and bad. She dies and is buried at the Bottom cemetery. Thereafter, Nel visits Eva Peace who is quite old and is kept in a nursing home where she tries to retell the moments she had together before Sula died. While walking home she begins to feel the loss of the relationship with the death of Sula, her single and true friend. Her judgment of Sula as bad begins to haunt her as she recalls an incident that happened when they were friends; the Chicken Little incident. Chicken little was a young boy who also lived at the Bottom. When playing on a tree, Sula lost grip and the child plunged in the river and drowned. They kept this secret for the whole of their lives. These are the memories that unsettled Nel emotionally, she begins to challenge the concepts of good and bad and the choices and sacrifices people make in life. Later on, she kills her drug addict sun as she struggles to raise his family single handedly. Sadly, we see her sitting and crying over Sulas grave at then cemetery as the story comes to an end. The story questions the decisions that people make in life; the calls for a rethinking of common societal problems. Critics mention the humor in the novel. For example the place called bottom is actually at the mountain top. The creation of binary oppositions in the novel not only makes it too interesting and informative but also lets the reader glance at the depth of common conventions in the community and the effect these societal conventions have on the lives of children who are supposed to be brought up in a comparatively free world where people are supposed to make free will. Satire as a theme is also developed in the story and it fuse well with the binary notion. The complexity of characters in the story conveys an account of human flaws in decision making and in the way we relate to other people in the community. While parents do everything in their power to ensure that their children justifiably lead a good life, little is done in ensuring that the children also get freedom. This makes the children not to acquire their own experiences in life but only proceed to copy the characters of their parents and grandparents. The fact that Sula turned out to be an exact replica of her mother and grandmother is an attestation to this fact. The book is feminine masterpiece and there is also an element of female chauvinism. This can be justified by the fact that all the major characters in the book are women and their actions are affected by men in a very little sense. Jude, Chicken Little and Nels son who became a drug addict after leaving the army are all victims of the women’s stereotyped dispositions. Shadrack being physically and emotionally scarred fro the experience of war is disowned and almost excommunicated by the community without the community fully understanding the reasons behind his behavior. Let us now give a critical analysis of Sula, the main character. Sula is the major influence on his friend Nel. Sula has been influenced by her mother leading to the passage of what can be argued to be a bad character trait through generations. Eva is a strong woman. She has endured loneliness, abject poverty but she is proud and unrelenting. Sula is quite an extraordinary woman, she is energetic but she has no suitable outlet to release her energies. This makes her potentially dangerous in that that she has no knowledge of wrongdoing. She does not understand that she sins when she sleeps with men and carelessly tosses them aside. Her sin is unintentional. The community, however do not understand the context of her upbringing or they simply do not care. They despise her but tolerate her. One very interesting aspect is that Sula is quite independent in her isolation, she does what she pleases and she simply does not care what everybody else does. She lives her the way it pleases her effectively earning her isolation from the community. After her death the rules shift suddenly, there is a renewed sense of acceptance, of defining right and wrong, good and bad. It is important to try and delve into the reasons why Nel and Sula united even after Sula betrayed their friendship by having an affair with Nels husband, Jude. Nel realizes that Sula was neither good nor evil; she was just indifferent to everything and everybody. Wives had to put extra effect in making sure that their husbands do not fall into bed with Sula, children were treated better so that they dont grow into adults who are indifferent and uncaring like Sula. This is good fortune to the bottom community because they were held together in the rethinking process. Remember the National Suicide day? After Sulas death the community did not have any productive outlet to channel their energies, they could not just sit and talk and so they become enraged and tear up the tunnel the result of unfocused energy. This eventual realization went a long way in ensuring that the residents of Bottom engaged in productive work for their survival. If Sula would have directed her energy to something else, maybe something a little more worthwhile would have resulted. Racial prejudice is highlighted when Jude is denied a chance to engage in the building of anew bridge, because of the color of his skin; he is denied the opportunity even though he is willing to do something productive. He stands in line for six days while the white boys get an opportunity to build the new River Road, his job at the hotel is demeaning and an insult to his masculinity. The scarcity of job opportunities remains to be the why Eva and Hannah had very little chance of gainful employment. They had to contend with the injustices being meted out on them because they were females and most importantly black. To escape the pangs of hunger in the winter, they have to prepare canned food in the summer. The life of Sula Peace, her childhood and her death in 1941is surrounded by the inability of the environment to shape her into a good woman The black community residing in Medallion; the bottom is judgmental but not in any way assisting to solve the crisis within itself. Sula’s life story shows us how the community and family can shape somebodys identity. She not only displays how children are nurtured but also her later relationship with the same community who natured her. She is a strong independent character but the community does not see her positive potentiality. To go against these societal norms she is not married and she sleeps around. The Peace family has been frowned upon by the Bottom community. This could have elicited an element of antagonism from Sula because she does not follow accepted societal conventions but in this case she is the protagonist and not the community. The novel poses her as the central character and the community constitutes only bits and pieces that seem to knit her life together in a predetermined fashion from the time of her birth to death. If change is to take place the community remains the playing field and Sula undoubtedly remains one of the players in the field. Whether she deserved to be rewarded for fearlessly instigating the rethinking process is not a matter of discussion as it is evident that her one and truly friend had in her own way knowingly or unknowingly lent credit to her stance in life by crying over her grave at the cemetery after realizing what Sula stood for. The character Sula is structure less, that any character interaction with her only invokes an element of structurelessness. This is because philosophically her evil is not her own but a reflection of what had been unknowingly imparted in her. Fortunately, her story is one where what is regarded as evil triggered change in the society. In analyzing one of the important masterpieces in the history of black American writing it is prudent to expunge on the growth of protagonism and non conformism in the society. The Work Of Milton Glaser Arts Essay Milton Glaser, was born in 1929 in New York, he attended the High School of Music Art. Known for his influential dynamic design, three of his most famous works include the I love New York logo, Bob Dylan poster and DC bullet logo, designed for DC Comics. Glaser has worked for a variety of agencies, designers, illustrators and co-founded Push Pin Studios in 1954. In 1977, Milton Glaser designed the I heart New York poster, unbeknown to him that it is this piece of design that would define his life. As a very simple piece of design to the eye, it holds a very clever message, Glaser explains how the design is to be translated to be fully understood, he says that, to understand the design you have to translate it. First of all you have to figure out that the I is a complete word, then you have to figure out that the heart is a symbol for an experience, then you have to figure out that NY are the initials for a place he then goes onto say that the issue in all communication is moving the brain, and puzzles move the brain, this then makes people feel good because they have solved the problem, just like this logo. The logo was designed to promote tourism in New York City and then became the trademark of the New York State. Glaser did the work pro bono as he only expected the campaign for more tourism to last a few months; never did he think it woul d have gone global, as this was not his original intention. However, the logo was a major success and has become so iconic worldwide and has been and still continues to be sold for years. The logo, then became furthermore prominent during the aftermath of the 911 terror attacks, as Glaser added a black smudge to the bottom corner of the heart symbolising a wound and a wounded heart is important to attain healing, and to acknowledge pain, despite this though the Department of Commerce were sceptic to adopt the creation which is why Glaser sent the new logo to New York Daily News, circulation close to a million copies around the city and the added the extra words more than ever, it helped create a sense of unity between the population and an awareness of support from visitors. Glaser felt that whilst working for Push Pin studios, that he became imprisoned by its reputation, and I wanted to try and invent a new ambiguous rep utation, leaving the agency to build up his own strong reputation. Glaser is a very ambitious individual, this is definitely something to idolize, not being scared or worried about other peoples views on what he produces or the context that surrounds it. Computers are to design as microwaves are to cooking, what a great statement! Design is an art whether a piece takes hours, days, months, weeks or even years, it is very precious. As a simple analogy, a microwave is an electrical appliance that uses technology to reheat and cook food; computers as technology lower the standard, classic knowledge as well as the craft of design. The definition of a designer seems to have become more blurred. The accessibility of technology now means that anyone with a computer and the correct software can become a designer. I believe in using a variety of styles, dependent upon which area of design Im working on and what purpose the design has. My styles seem to have become more varied in the last couple of years, which is great, becoming too rounded on a particular style is good, if wanting to familiarize people to easily recognise a defined style and the designer, but then having that comfort of working in one style I feel makes people become conformed to that one and only style, not allowing any room for change, just how us humans work in general, we dont like change, having a variety of rounded styles is great and something I definitely believe in. When I work on HTML and websites, I really do prefer to be minimalistic, ordered and very structured enabling a good usability for browsers, yet this may be seen more for purpose than design as I do not like over complex websites that confuse the browser. However, when designing a piece of work purely around fine art, my thought is exactly the opposite, I adore being slightly abstract and bright, bold colours. I do this as I find art truly is a form of expression and communication, just how Glaser states that communication is all about moving the brain, which is done through puzzles, so when someone can identify what a piece of work is and they feel good they have done this, it shows there is effective communication and expression in a piece of work. Typography is not really one of my strong suits, but I really do have to acknowledge the importance of its use amongst design. It is typically questioned whether there are currently too many fonts, as there are hundreds and possibly thousands of websites that offer free fonts for download. I disagree with the statement that there are too many fonts, as it really is the understanding of the fonts that is lacked these days. Having this knowledge can really define the difference between a good and excellent designer I feel. Understanding and knowing the main fonts, font families and the use of line height, kerning and tracking is very important in any area of design, if a designer is not aware of any of these aspects of typography, knowing when and how to use them may become ineffective, as the art of typography is to communicate efficiently and therefore has be clean, legible and well designed, knowing the basics is essential. I really aspire to travel wherever design takes me, and experience a bit of everything. I have my strong suits and my weak suits, but there is so much I want to learn. I am very good at fine art practice and thoroughly enjoy this; this is because it is a very way for me to communicate with people without using words. As well as this, the amount of historical and current influences that inspire my work is vast, I enjoy combining influences to create a mixed piece of design, this may be seen as unconventional and wrong but the possibilities within design are endless, as Glaser states my sense is not to be allergic to style or mores in time, just my thoughts exactly, I really enjoy combining styles and references. THE BALLARD FUEL CELL The Ballard fuel cell is a power generating device which combines hydrogen (which can be obtained from methanol, natural gas, petroleum) and oxygen without the use of combustion in order to generate electricity. Since fuel cells operate very quietly and efficiently and their only emissions are pure water and heat they are expected to be the future of power generating machines. The PEM fuel cell is made of two plates with a plastic membrane coated with a catalyst in the middle. Hydrogen is then fed through the channels on one side of the plates and oxygen is fed through the other side. The hydrogen and oxygen atoms are drawn toward each other. Only one part of the hydrogen atom – the proton – can pass through the membrane. The electron has to take the long way around through an external circuit. This creates electricity. The oxygen side attracts protons and electrons that have traveled through the external circuit. This is where the byproducts water and heat are formed. Each fuel cell operates continuously as long as hydrogen is supplied. Single cells are combined to produce the required amount of power. PEM fuel cells operate at around 90 degrees Celsius and give off 90 degrees Celsius of heat, which is a fairly low temperature. This is much better than high temperature combustion engines, which operate at about 2500 degrees Celsius and give off 125 degrees Celsius of heat. This means the Ballard fuel cell can react quickly to load changes and makes it ideal for motor vehicles.
